N3C Partners with AIM-AHEAD on a NCATS Real-World Data Training Program


The AIM-AHEAD Consortium, which strives to enhance the participation and representation of researchers and communities currently underrepresented in the development of AI/ML models, has recently partnered with NCATS to develop and offer a real-world data training program using N3C resources.


The program, now underway, offers trainees hands-on experience in real-world data analysis with N3C infrastructure, good algorithmic practice, and clinical data management. The Guide to N3C  features as part of the instructional materials, and members of the N3C Logic Liaisons, Education and Training Domain Team, and Good Algorithmic Practice teams (Johanna Loomba, Shawn O'Neil, and David Sahner) have contributed to design and instruction. This effort is also piloting new education-focused resources in N3C that will become more broadly available as part of the N3C Clinical initiative. Life changed for María Patrizia Santos during a visit to Peru in 2017. It was not from a mystical experience at Machu Picchu in the Andes or one of the South American country's many other ancient sites, but from interviewing more than 150 caregivers about childhood anemia in the southern city of Arequipa.

Infrastructure, consultation for multi-site studies using electronic health data

NIH-funded Observational Health Data Sciences Initiative links over 3,000 collaborators across approximately 80 countries with electronic health records data repositories that collectively contain over 900 million unique patient records.

Association of Premorbid GLP-1RA and SGLT-2i Prescription Alone and in Combination with COVID-19 Severity


People with type 2 diabetes are at heightened risk for severe outcomes related to COVID-19 infection, including hospitalization, intensive care unit admission, and mortality. This study was designed to examine the impact of premorbid use of glucagon-like peptide-1 receptor agonist (GLP-1RA) monotherapy, sodium-glucose cotransporter-2 inhibitor (SGLT-2i) monotherapy, and concomitant GLP1-RA/SGLT-2i therapy on the severity of outcomes in individuals with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection.

Privacy-preserving record linkage across disparate institutions and datasets to enable a learning health system: The national COVID cohort collaborative (N3C) experience


Research driven by real-world clinical data is increasingly vital to enabling learning health systems, but integrating such data from across disparate health systems is challenging. As part of the NCATS National COVID Cohort Collaborative (N3C), the N3C Data Enclave was established as a centralized repository of deidentified and harmonized COVID-19 patient data from institutions across the US. However, making this data most useful for research requires linking it with information such as mortality data, images, and viral variants. The objective of this project was to establish privacy-preserving record linkage (PPRL) methods to ensure that patient-level EHR data remains secure and private when governance-approved linkages with other datasets occur.

Pregnancy Outcomes among Pregnant Persons after COVID-19 Vaccination: Assessing Vaccine Safety in Retrospective Cohort Analysis of U.S. National COVID Cohort Collaborative (N3C)


COVID-19 vaccines have been shown to be effective in preventing severe illness, including among pregnant persons. The vaccines appear to be safe in pregnancy, supporting a continuously favorable overall risk/benefit profile, though supportive data for the U.S. over different periods of variant predominance are lacking. 

The Intersections of COVID-19, HIV, and Race/Ethnicity: Machine Learning Methods to Identify and Model Risk Factors for Severe COVID-19 in a Large U.S. National Dataset


We investigate risk factors for severe COVID-19 in persons living with HIV (PWH), including among racialized PWH, using the U.S. population-sampled National COVID Cohort Collaborative (N3C) data released from January 1, 2020 to October 10, 2022. We defined severe COVID-19 as hospitalized with invasive mechanical ventilation, extracorporeal membrane oxygenation, discharge to hospice or death.

The National COVID Cohort Collaborative (N3C) is a complementary and synergistic partnership among the Clinical and Translational Science Awards (CTSA) Program hubs, the National Center for Data to Health (CD2H), distributed clinical data networks (PCORnet, OHDSI, ACT, TriNetX), and other partner organizations, with overall stewardship by NIH’s National Center for Advancing Translational Sciences (NCATS). The N3C aims to improve the efficiency and accessibility of analyses using a very large row-level (patient-level) COVID-19 clinical dataset, demonstrate a novel approach for collaborative pandemic data sharing, and speed understanding of and treatments for COVID-19.
